[目的]探讨枳实总黄酮苷提取物(aurantii fructus immaturus flavonoid,AFIF)对功能性消化不良(functional dyspepsia,FD)模型大鼠胃适应性的影响及其可能的作用途径。[方法]10日龄雄性SD大鼠86只,随机分为正常组22只及造模组64只,造模组采用碘乙酰胺(iodoacetamide,IA)灌胃法联合夹尾应激法诱导大鼠FD模型;至大鼠8周龄时将造模组大鼠随机分为枳实总黄酮苷提取物大、中、小剂量组及模型组,分别予大、中、小剂量的枳实总黄酮苷提取物或生理盐水干预14d,给药结束后,各组大鼠经食道置入胃内气囊,应用电子恒压器对胃内气囊分阶段加压,检测大鼠胃适应性,加压程序为:以1mmHg为起始压力,维持10min后,每分钟压力升高1mmHg直至10mmHg(斜坡期),在10mmHg压力下维持10min(平台期)。应用离体平滑肌张力测定技术检测不同浓度的枳实总黄酮苷提取物对大鼠近端胃体离体纵行平滑肌张力的影响。[结果]近端胃适应性检测结果显示:在斜坡期及平台期,模型组大鼠胃适应性较正常组显著降低(P<0.05);枳实总黄酮苷提取物大剂量组(大剂量组)大鼠胃适应性在斜坡期及平台期均较模型组显著升高(P<0.05);枳实总黄酮苷提取物中剂量组(中剂量组)大鼠胃适应性在平台期较模型组显著升高(P<0.05);枳实总黄酮苷提取物小剂量组(小剂量组)大鼠胃适应性较模型组差异无统计学意义。近端胃体离体平滑肌张力测定结果显示:近端胃体纵行平滑肌张力随枳实总黄酮苷提取物浓度升高而降低,当枳实总黄酮苷提取物浓度达到6 000μg/ml及10 000μg/ml时具有统计学意义(P<0.05)。[结论]枳实总黄酮苷提取物对功能性消化不良大鼠近端胃适应性具有一定的调节作用,其作用途径可能与降低近端胃体平滑肌张力有关。
[Objective] To investigate the effect of aurantii fructus immaturus flavonoid (AFIF) on gastric responsiveness in rats with functional dyspepsia (FD) and its possible pathways. [Methods] A total of 86 male 10-day-old male Sprague-Dawley rats were randomly divided into normal control group (n = 22) and model group (n = 64). The rats in model group were treated with iodoacetamide Rats FD model; to rats at 8 weeks of age, the rats in the model group were randomly divided into large, medium and low dose of Fructus Aurantii Immaturus extract group and model group, respectively, to large, medium and small doses of the total citrus aurantium Flavonoid glycosides extract or saline intervention 14d, after the end of administration, rats in each group were placed in the stomach intragastric air bags, the application of electronic pressure on the gastric balloon pressure in stages, the gastric adaptation test, pressure The procedure is: Starting at 1 mmHg and maintaining for 10 min, the pressure is increased by 1 mmHg per minute up to 10 mmHg (ramp) and maintained at 10 mmHg for 10 min (plateau). The effects of different concentrations of total flavonoid glycoside extract on tension of isolated longitudinal smooth muscle of rat proximal gastric body were detected by in vitro smooth muscle tension measurement. [Result] The results of proximal gastric adaptation test showed that gastric adaptation in model group was significantly lower than that in normal group (P <0.05) at the slope stage and plateau stage. The high dose group (P <0.05). The stomach adaptability of middle dose group (middle dosage group) of total flavonoid glycoside extract of Fructus Aurantii Immaturus was better than that of the model group in the plateau stage (P <0.05). There was no significant difference in gastric adaptation between low dose group (low dose group) and model group (P <0.05). The results of in vitro smooth muscle tension of proximal gastric body showed that the longitudinal smooth muscle tension of proximal gastric body decreased with the increase of the concentration of total flavonoid glycosides in Fructus Aurantii Immaturus. When the concentration of total flavonoid glycosides in Fructus Aurantii Immaturus reached 6 000μg / ml and 10 000μg / ml was statistically significant (P <0.05). [Conclusion] The total flavonoid glycoside extract of Fructus Aurantii Immaturus has some regulative effects on proximal gastric adaptation of functional dyspepsia rats, which may be related to decreasing the tension of proximal gastric smooth muscle.
