蜂斗菜(Petasites Japonicus Fr.Schmidt)是雌雄异株作物,通常只种植雌株,用地下茎繁殖,再加上三倍体栽培种的不孕性(2 n=87,x=29),进行杂交育种极其困难.本文研究了蜂斗菜叶肉原生质体的分离和培养,并讨论了在育种上利用的可能性. 取供试品种“Aichi-Wase-buki”生长的地下茎15~20 cm进行表面消毒,首先插入70%酒精60 S,再浸入有效氯为0.5%的漂白粉溶液10min.消毒后,在解剖镜下剥取带有叶原基的茎尖(0.5~1.1mm)接种于Ms+BA 2 mg/l的培养基上.待茎叶伸出后,切割移植在Ms+BA
