目的:探讨化痰活血益气方的抗心肌肥厚作用及对心肌间质Ⅰ、Ⅲ型胶原的影响。方法:采用异丙肾上腺素诱导的大鼠心肌肥厚模型(1 mg/kg,sc,连续10 d),48只大鼠随机分为6组:正常组、模型组、化痰活血益气方组(4.88 g/kg、9.77 g/kg、19.53 g/kg)和卡托普利组(0.02 g/kg)。14 d相应药物干预后,测定左心室重量指数(left ventricle mass index,LVMI)、全心重量指数(heart mass index,HMI),行HE染色测量心肌细胞表面积、行Masson染色测量胶原容积分数(collagen volume fraction,CVF),免疫组化法检测Ⅰ、Ⅲ型胶原表达,实时荧光定量RT-PCR检测Ⅰ、Ⅲ型胶原mRNA的表达,Western blot检测Ⅰ、Ⅲ型胶原蛋白的表达。结果:与正常组比较,模型组的LVMI、HMI、心肌细胞表面积、CVF以及Ⅰ、Ⅲ型胶原在形态学、mRNA、蛋白质层面的表达均明显升高;与模型组比较,化痰活血益气方组(4.88 g/kg、9.77 g/kg、19.53 g/kg)和卡托普利组的LVMI、HMI、心肌细胞表面积、CVF以及Ⅰ、Ⅲ型胶原在形态学、mRNA、蛋白质层面的表达均有不同程度的降低,化痰活血益气方9.77 g/kg组的上述各项指标始终存在显著性差异。结论:化痰活血益气方可以有效改善异丙肾上腺素诱导的心肌肥厚并降低Ⅰ、Ⅲ型胶原的表达。
Objective: To investigate the anti-hypertrophy effect of Huatan Huoxue Yiqi Prescription and its effect on type I and type III collagen in myocardial interstitium. Methods: The rat model of myocardial hypertrophy was induced by isoprenaline (1 mg / kg, sc for 10 days). Forty-eight rats were randomly divided into 6 groups: normal group, model group, (4.88 g / kg, 9.77 g / kg, 19.53 g / kg) and captopril (0.02 g / kg). The left ventricle mass index (LVMI) and heart mass index (HMI) were measured after 14 days of drug intervention. The myocardial cell surface area was measured by HE staining and the collagen content (collagen) (CVF). The expressions of type I and type III collagen were detected by immunohistochemistry. The expressions of type I and type III collagen mRNA were detected by real-time fluorescence quantitative RT-PCR. The expressions of type I and type III collagen were detected by Western blot. Results: Compared with the normal group, the expression of LVMI, HMI, myocardial cell surface area, CVF and type Ⅰ and type Ⅲ collagen in the model group were significantly increased at the morphological, mRNA and protein levels. Compared with the model group, LVMI, HMI, cardiomyocyte surface area, CVF and the expression of type I and type III collagen in the four groups (4.88 g / kg, 9.77 g / kg, 19.53 g / kg) and captopril group at the morphological, mRNA and protein levels All had different degrees of reduction. There were always significant differences among the above indexes in the group of 9.77 g / kg of Huatan Huoxue Yiqi Prescription. Conclusion: Huatan Huoxue Yiqi Prescription can effectively improve isoproterenol-induced cardiac hypertrophy and reduce the expression of type I and type III collagen.
