目的用生物测定法考察肉苁蓉多糖(CDPS)在Caco-2细胞模型的吸收转运。方法用培养于Transwell上的Caco-2细胞模型研究CDPS的吸收转运;采用生物活性测定法测定CDPS在接受侧介质中的含量。结果Caco-2细胞单层极性与完整性符合实验要求,CDPS在1.87~40mg·mL-1内,与RAW264.7细胞NO分泌呈明显量效关系,转运量随供侧(AP侧)浓度的增加而增大,不同浓度CDPSPapp(A-B)均小于1×10-7cm.s-1。结论采用生物活性测定CDPS的Papp具可行性,该药属吸收不良药物,口服吸收率应小于1%。
Objective To investigate the absorption and transport of Cistanche deserticola polysaccharide (CDPS) in Caco-2 cell model by bioassay. Methods The Caco-2 cell model cultured on Transwell was used to study the absorption and transport of CDPS. The bioactivity assay was used to determine the content of CDPS in the medium of the recipient. Results The polarity and integrity of monolayer of Caco-2 cells accorded with the experimental requirements. CDPS was 1.87 ~ 40 mg · mL-1 and showed a significant dose-effect relationship with NO production in RAW264.7 cells. Of CDPSPapp (AB) were less than 1 × 10-7cm · s-1. Conclusion The feasibility of using Papp as a bioassay for the determination of CDPS is poorly absorbed and the oral absorption rate should be less than 1%.
