目的合成3β-羟基-5-雄甾烯-17-酮-丁二酸酯(5-烯雄甾酯)人工抗原。方法以N,N-二环己基碳二亚胺和N-羟基琥珀酰亚胺为催化剂,采用活性酯法将5-烯雄甾酯与牛血清白蛋白偶联。结果产物经元素分析、质谱和红外光谱确定,合成得到每个BSA载体连接6个半抗原小分子的人工抗原。结论采用琥珀酸酐衍生化5-烯雄甾酯,活性酯法合成抗原,产率高、副产物少。
Objective To synthesize 3β-hydroxy-5-androstene-17-one-succinate (5-androstane ester) artificial antigen. Methods N, N-dicyclohexylcarbodiimide and N-hydroxysuccinimide were used to catalyze the coupling of 5-ene androstane with bovine serum albumin by the active ester method. Results The products were identified by elemental analysis, mass spectrometry and infrared spectroscopy. The artificial antigens were obtained by connecting six small hapten molecules to each BSA vector. Conclusion Succinic anhydride derivatization of 5-androstane esters, active ester synthesis of antigens, high yield, fewer by-products.
