目的:探讨丙酮固定骨髓标本乙二醇-甲基丙烯酸酯(glycol-methacrylate,GMA)塑胶包埋(塑包)切片内保留敏感造血分化抗原的新方法。方法:使用冷丙酮固定结合简易三步塑包体系,观察52例血液病患者骨髓GMA包埋切片内14种单克隆标志物的表达情况。结果:简易三步塑胶包埋体系不仅能提供良好的细胞形态,细胞抗原性也获较佳保存,并可在切片内进行c-myc原癌基因表达的检测。结论:建立了一种骨髓活检的简易塑包操作技术,既可保留良好的抗原性,也有助于观察免疫组织化学标志物相关的形态学细节。
Objective: To explore a new method for preserving sensitive hematopoietic differentiation antigen in plastic-embedded (plastic bag) sections of acetone-fixed bone marrow specimens. Methods: The expression of 14 kinds of monoclonal markers in the bone marrow GMA embedded sections of 52 patients with hematological diseases was observed with cold acetone fixation and simple three-step plastic bag system. Results: The simple three-step plastic-embedded system could not only provide good cell morphology, but also preserve the antigenicity of cells and detect the expression of c-myc oncogene in the sections. CONCLUSIONS: A simple plastic bag manipulating technique for bone marrow biopsy was established that retains good antigenicity as well as observes the morphological details associated with immunohistochemical markers.
