HPLC法检测鸡蛋中DHA-EE和EPA-EE含量的条件优化

为利用高效液相色谱法检测衍生化鸡蛋样品中的二十碳五烯酸乙酯(EPA-EE)和二十二碳六烯酸乙酯(DHA-EE),使用super C18色谱柱,考察了流动相有机相比例、流速及柱温改变对EPA-EE和DHA-EE分离的影响,确定了等度洗脱、流动相为甲醇-水(95∶5,Ⅵ V)、流速1.0 mL/min、柱温30℃、检测波长220 nm的色谱条件.该方法下,EPA-EE和DHA-EE分别在线性范围内浓度和峰面积呈现良好的线性关系,相关系数r分别为0.999 2和0.999 0,采用上述方法测定了ω-溴苯乙酮衍生化的鸡蛋中混合脂肪酸,结果表明EPA-EE和DHA-EE二峰可达到基线分离.本方法快速、简单、重复性好,为鸡蛋中多不饱和脂肪酸含量的快速检测提供了思路.
