目的本文筛选CCl4致SD大鼠急性肝损伤模型的最佳给药浓度和剂量,追踪观察模型大鼠肝损伤后修复过程。方法本文采用随机对照分组的研究方法,对不同的SD大鼠模型组给予不同剂量的CCl4一次性腹腔注射。给药后48 h检测大鼠血清ALT, AST。各组随机处死一只SD大鼠,取肝组织制成病理切片镜下观察。追踪观察筛选后的CCl4模型大鼠肝组织病理变化恢复情况。结果本实验发现3A-7A组的ALT,AST较对照组明显升高(P<0.05或P<0.01),随着CCl4的剂量的增加,大鼠肝组织的病理损伤加重,6A和7A组可见自然死亡大鼠。结论本试验筛选了CCl4致SD大鼠急性肝损伤模型的较好CCl4的剂量。该模型肝损伤明显,死亡率低,对其他重要脏器影响小,可作为急性肝损伤研究的较好模型。
Objective To screen the optimal concentration and dose of CCl4-induced acute liver injury in SD rats and observe the repair process after liver injury in model rats. Methods In this study, a randomized controlled study was conducted in which different doses of CCl4 were intraperitoneally injected into different SD rat models. At 48 h after administration, serum ALT and AST were measured. One SD rat was randomly sacrificed in each group, and the liver tissue was taken for pathological observation. Tracing and observing the pathological changes recovery of liver tissue of CCl4 rat after screening. Results The level of ALT and AST in 3A-7A group was significantly higher than that in control group (P <0.05 or P <0.01). With the increase of CCl4 dose, the pathological damage of liver tissue in rats increased. Naturally dead rats. Conclusion This experiment screened the CCl4-induced acute liver injury model of CCl4 better CCl4 dose. This model has obvious hepatic injury, low mortality and little effect on other important organs, which can be used as a good model for the study of acute liver injury.
