目的采用一个简单的、实用的方法评价临床个体患儿体内CYP3A酶活性;预测儿童白血病患者体内依托泊苷血药浓度、药动学参数。方法采用HPLC测定药物浓度,一点法计算药物消除半衰期,尿样法测定体内CYP3A酶活性。结果儿童白血病患儿体内依托泊苷药物浓度与CYP3A酶活性相关性较差,而其t1/2与个体在化疗前CYP3A酶活性变化的相关性较好。结论依托泊苷t1/2与儿童白血病患儿体内CYP3A酶活性有一定的相关性。
Objective To evaluate the activity of CYP3A in clinical children with a simple and practical method. Predict the concentration of etoposide in children with leukemia and the pharmacokinetic parameters. Methods The drug concentration was determined by HPLC, the half-life of drug elimination was calculated by one-point method, and the CYP3A enzyme activity was measured by urine sample. Results The correlation between the drug concentration of etoposide and the activity of CYP3A in children with childhood leukemia was poor, and the correlation between t1 / 2 and the change of CYP3A activity before chemotherapy in children with childhood leukemia was good. Conclusion Etoposide t1 / 2 is associated with CYP3A activity in children with childhood leukemia.
