目的:研究黄根东莨菪内酯抗大鼠肝纤维化作用及其作用机制。方法:采用复合因素造模制备肝纤维化大鼠模型。造模四周后,灌胃给予东莨菪内酯两周,测定肝功能指标(AST、ALT、TBIL、A/G比值)及肝纤维化指标(透明质酸(HA)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ)、层粘蛋白(LN));定量分析相关细胞因子(TGF-β1、TNF-α、IFN-α和HGF)的表达;通过mssson染色法观察大鼠肝纤维化病理变化。结果:东莨菪内酯组(2.27×10~(-2)mg/kg)和东莨菪内酯组(7.56×10~(-3)mg/kg)均可显著性降低AST、ALT活性,东莨菪内酯组(2.27×10~(-2)mg/kg)可明显降低TBIL活性,使PCⅢ和α-SMA水平下降,并显著性降低TGF-β1和TNF-α含量的同时,升高HGF水平;东莨菪内酯组(7.56×10~(-3)mg/kg)可显著性降低PCⅢ和Ⅳ胶原水平,降低α-SMA表达,既可显著性降低TGF-β1和TNF-α含量,又可显著性升高IFN-α和HGF水平;东莨菪内酯给药组可减轻肝纤维化大鼠病理损伤程度。结论:从黄根中提取出的东莨菪内酯能保护肝纤维化大鼠肝脏,降低肝脏细胞外基质的水平,正向调控与肝纤维相关细胞因子,改善大鼠肝纤维化病变过程,具有抗肝纤维化作用。
OBJECTIVE: To study the anti-hepatic fibrosis effect of flavonoids from Rhizoma from root and its mechanism. Methods: The hepatic fibrosis rat model was made by compounding factors. Four weeks after modeling, scopoletin was orally administered for two weeks. The indexes of liver function (AST, ALT, TBIL and A / G) and the indexes of liver fibrosis (HA, PCⅢ (Ⅳ) and laminin (LN)). The expression of related cytokines (TGF-β1, TNF-α, IFN-α and HGF) was quantitatively analyzed. The liver fibrosis was observed by mssson staining Pathological changes. Results: The activities of AST and ALT were significantly lower in the scopoletin group (2.27 × 10 -2 mg / kg) and the scopoletin group (7.56 × 10 -3 mg / kg) (2.27 × 10 ~ (-2) mg / kg) could significantly decrease the activity of TBIL, decrease the levels of PCⅢ and α-SMA, and significantly reduce the content of TGF-β1 and TNF-α (7.56 × 10 ~ (-3) mg / kg) could significantly reduce the levels of PC Ⅲ and Ⅳ collagen and decrease the expression of α-SMA, which could significantly reduce the levels of TGF-β1 and TNF-α, But also significantly increased the level of IFN-a and HGF; scopoletin administration group can reduce the degree of pathological damage in rats with hepatic fibrosis. Conclusion: Scopoletin extracted from Radix Gentianae can protect the liver of rats with hepatic fibrosis and reduce the level of extracellular matrix in the liver, positively regulate the cytokines associated with liver fibrosis, improve the pathological changes of liver fibrosis in rats, Fibrosis.
