目的:分离克隆测序胃癌高表达基因gcys18 的部分c D N A. 方法:采用m R N A 差异展示技术从胃癌 G C7901 与胃粘膜 G E S1 细胞株中分离出一差异表达基因片段,对其命名,并进行克隆、测序及 Gen Bank 检索;以此片段作探针与细胞株总 R N A 进行打点杂交. 结果:从差示 P C R 电泳凝胶中分离出大小为416 bp 的差异表达片段,命名为gcys18; R N A 打点杂交证实gcys18 在 G C7901 中呈高表达,在 G E S1 中呈低表达,表明gcys18 是真正差异表达片段;对gcys18 进行了序列分析及 Gen Bank 检索,示其为新序列后递交给 Gen Bank,它的 Gen Bank 接受号为 A F071057. 结论:分离并克隆了胃癌高表达基因gcys18 的部分c D N A,此基因可能与胃癌发生发展有关.
OBJECTIVE: To isolate and sequence partial c D N A gene of gastric cancer highly expressed gene gcys18. METHODS: A differentially expressed gene fragment was isolated from gastric cancer G C7901 and gastric mucosae G E S 1 cell strain by m R N A differential display technology, and was named, cloned, sequenced, and retrieved by GenBank; As a probe and cell line total R N A for spot hybridization. RESULTS: A 416 bp differentially expressed fragment was isolated from the differential P C R electrophoresis gel and named gcys18. Rn A spot hybridization confirmed the high expression of gcys18 in G C7901 in G E S . Low expression in 1 indicates that gcys18 is a truly differentially expressed fragment; sequence analysis of gcys18 and GenBank search are presented as a new sequence and submitted to Gen Bank. Its Gen Bank acceptance number is A F071057 . CONCLUSION: A portion of c D N A gene of gcys18, a highly expressed gastric cancer gene, was isolated and cloned. This gene may be involved in the development of gastric cancer.
