目的:从行为学角度,观察柚皮苷对转基因果蝇阿尔茨海默病(AD)模型的影响。方法:采用Gal4/USA系统建立Aβ果蝇模型;连续给药后进行行为学实验,观察柚皮苷对AD果蝇寿命的影响。对照组和模型组培养基中加入等容积培养菌(酵母菌)后,给药组分别给予20mmol/L和50mmol/L柚皮苷,抑制剂组分别给予20mmol/L柚皮苷、20mmol/L L-NAME和50mmol/L柚皮苷、40mmol/L L-NAME。通过爬行实验观察柚皮苷对阿尔茨海默症果蝇寿命的影响。结果:与对照组比较,模型组果蝇寿命明显减少;与模型组相比,柚皮苷治疗组能明显增加果蝇寿命;与治疗组相比,抑制剂组能显著减少果蝇寿命。结论:柚皮苷对AD果蝇模型寿命具有保护作用,其作用机制可能与其激活e NOS表达有关。
Objective: To observe the effect of naringin on the Alzheimer’s disease (AD) model of transgenic Drosophila from behavioral perspective. Methods: The Aβ fruit fly model was established by using Gal4 / USA system. After continuous administration, behavioral tests were conducted to observe the effect of naringin on the lifespan of AD fruit flies. After adding equal volume of culture medium (yeast) to the culture medium of the control group and the model group, the naringin of 20 mmol / L and 50 mmol / L were given to the treatment group, and the naringin of 20 mmol / L and 20 mmol / L L-NAME and 50mmol / L naringin, 40mmol / L L-NAME. The effects of naringin on the life span of Drosophila melanogaster in Alzheimer’s disease were observed by the creeping experiment. Results: Compared with the control group, the life span of the fruit flies in the model group was significantly reduced. Compared with the model group, the naringin treatment group significantly increased the life span of the fruit flies. Compared with the treatment group, the inhibitor group significantly reduced the life span of the fruit flies. Conclusion: Naringin has a protective effect on the lifespan of AD flies, and its mechanism may be related to the activation of eNOS expression.
