增生性瘢痕表皮细胞双向电泳图谱的建立及差异蛋白展示

目的建立在体增生性瘢痕表皮细胞蛋白质分子双向电泳(2-DE)技术,筛选出增生性瘢痕表皮细胞较正常表皮细胞差异表达的蛋白质,为揭示表皮细胞在增生性瘢痕形成过程中的作用奠定基础.方法利用消化法分离出在体的增生性瘢痕组织中的表皮细胞,提取其中总蛋白质,利用双向电泳技术展示蛋白质分子的表达,通过Melanie 3.0软件对凝胶图像进行分析,将结果与数据库中正常表皮细胞2-DE图谱进行比较.结果建立了能够显示超过600个蛋白质点的在体增生性瘢痕表皮细胞双向电泳图谱,通过比较筛选出在位置、形状或密度上存在差异的蛋白质点24个.其中高表达的点有8个,低表达点有9个,表达缺失或极低表达点有4个,新发现蛋白质点3个.结论采用消化法分离在体增生性瘢痕表皮细胞建立其2-DE图谱的方法是可行的,为进一步进行增生性瘢痕相关性蛋白质组研究奠定了基础.初步筛选出的24个差异蛋白质点提示增生性瘢痕表皮细胞的异常可能与增生性瘢痕的形成有关.
