反义寡核苷酸可以特异地抑制基因表达 ,设计应用与 bcr-abl融合基因m RNA互补的反义寡核苷酸作用于 K562 细胞 ,结果显示 :反义寡核苷酸对 K562 细胞的生长有明显的抑制作用 ,硫代化与未修饰反义寡核苷酸相比有明显的稳定性 ;反义寡核苷酸与VP16联合应用 ,可明显提高对 K562 细胞的增殖抑制作用。反义寡核苷酸的应用将为慢性粒细胞白血病患者自体骨髓移植体外净化提供新的途径
Antisense oligonucleotides can specifically inhibit the gene expression. Antisense oligonucleotides complementary to the bcr-abl fusion gene m RNA were designed to act on K562 cells. The results showed that antisense oligodeoxynucleotides could inhibit the growth of K562 cells The antisense oligodeoxynucleotides combined with VP16 could obviously inhibit the proliferation of K562 cells. The application of antisense oligonucleotides will provide a new way for in vitro purification of autologous bone marrow transplantation in patients with chronic myeloid leukemia
