为探讨外周血淋巴细胞中金属硫蛋白基因表达作为对镉易感性生物标志物的可能性 ,采用分离人外周血淋巴细胞(PBL) ,体外培养 ,培养液中加入镉 ,检测接触镉后的细胞存活率、DNA、单链损伤、乳酸脱氢酶和金属硫蛋白 (MT)基因表达。结果 :体外接触 1 0 μmol/LCdCl2 6小时 ,细胞存活率在吸烟组和不吸烟组间无显著性差异。MT基因基础表达、诱导表达和诱导能力在组间未见差异。PBL中MT基础表达与LDH泄漏、彗星率和血镉水平呈显著相关 ,和细胞存活率则无相关。MT诱导能力与LDH泄漏和彗星率改变呈负相关。随MT基因诱导能力的上升 ,LDH泄漏和彗星率明显降低。提示PBL中MT基因诱导能力与其对镉的易感性有关 ,初步提示其作为镉接触的易感性标志物的可能性。
To investigate the possibility of metallothionein gene expression in peripheral blood lymphocytes as a biomarker of susceptibility to cadmium, human peripheral blood lymphocytes (PBLs) were isolated from human peripheral blood lymphocytes and cultured in vitro. Cadmium Survival, DNA, single-stranded damage, lactate dehydrogenase and metallothionein (MT) gene expression. Results: In vitro exposure to 10 μmol / L CdCl2 for 6 hours showed no significant difference in cell viability between smokers and nonsmokers. The basic expression of MT gene, induction of expression and induction ability in the group did not differ. The basal expression of MT in PBL was significantly correlated with LDH leakage, comet rate and blood cadmium levels, but no correlation with cell survival rate. MT induction was negatively correlated with LDH leakage and change of comet rate. With the increase of the inductivity of MT gene, LDH leakage and comet rate decreased significantly. These results suggest that the induction of MT gene in PBL is related to its susceptibility to cadmium, suggesting that it may be a potential marker of susceptibility to cadmium exposure.
