目的:观察卡维地洛对心肌梗死后大鼠转化生长因子-β1和Ⅰ型胶原表达的影响。方法:结扎左冠状动脉前降支建立大鼠急性心肌梗死模型,心肌梗死后存活24 h者32只随机分为2组:观察组给予卡维地洛10 mg/(kg.d);对照组仅给予相同体积的生理盐水15 mL/(kg.d);假手术组10只为仅穿线不结扎左前降支者,给予生理盐水15 mL/(kg.d),均经灌胃给药。观察各组4周末血流动力学和心肌组织中转化生长因子-β1和Ⅰ型胶原的变化。结果:观察组与假手术组比较,4周末大鼠心脏质量、左心室质量、心脏质量指数、左心室质量指数均增加(P<0.05);与对照组比较,上述指标下降,差异有统计学意义(P<0.05);观察组大鼠血流动力学指标及心室重塑指标均显著改善。结论:卡维地洛降低转化生长因子-β1的表达和抑制Ⅰ型胶原的沉积,从而改善心肌梗死后心室重塑。
Objective: To observe the effect of carvedilol on the expression of transforming growth factor-β1 and type Ⅰ collagen in rats after myocardial infarction. Methods: Acute myocardial infarction model was established by ligation of the left anterior descending coronary artery. Thirty-two animals survived 24 hours after myocardial infarction were randomly divided into 2 groups: the control group received carvedilol 10 mg / (kg · d) Only given the same volume of saline 15 mL / (kg · d); sham-operated group of 10 only threading without ligation of the left anterior descending artery, given saline 15 mL / (kg.d), were given by gavage. The changes of hemodynamics and the levels of transforming growth factor-β1 and type Ⅰ collagen in myocardium were observed at the 4th weekend in each group. Results: Compared with the sham operation group, the heart mass, left ventricular mass, cardiac mass index and left ventricular mass index increased in the observation group and the sham operation group at 4 weeks (P <0.05). Compared with the control group, the above indexes decreased (P <0.05). The indexes of hemodynamics and ventricular remodeling in observation group were significantly improved. Conclusion: Carvedilol can reduce the expression of TGF-β1 and inhibit the deposition of type Ⅰ collagen, so as to improve ventricular remodeling after myocardial infarction.
