目的:测定慈姑粗多糖中游离蛋白和结合蛋白的含量。方法:用水提醇沉法提取慈姑中的水溶性多糖,经Sevag法将粗多糖脱去蛋白得精制多糖;用微量凯氏定氮法测定粗多糖和精制多糖中蛋白质含量。结果:经水提醇沉法所得的慈姑粗多糖中蛋白质含量为5.80%,慈姑精制多糖中蛋白质含量为2.67%,是因为Sevag法适用于游离蛋白的去除,但无法除去多糖中的结合蛋白。结论:慈姑多糖中含3.13%游离蛋白和2.67%结合蛋白。
Objective: To determine the content of free protein and binding protein in arrowhead polysaccharide. Methods: Water-soluble alcohol was used to extract water-soluble polysaccharides from Hericium auricula. The crude polysaccharides were deproteinized by Sevag method to obtain polysaccharides. The contents of crude polysaccharides and refined polysaccharides were determined by micro-Kjeldahl method. Results: The protein content of Cruciferae polysaccharide was 5.80% and the protein content of Cretaceous polysaccharide was 2.67%. The reason was that Sevag method was suitable for the removal of free protein but could not remove the binding protein from polysaccharide. Conclusion: Sagittaria polysaccharide contains 3.13% free protein and 2.67% binding protein.
