目的 为了通过细胞融合研制肺癌疫苗,需要建立有适合细胞融合的肺癌抗原细胞。方法 用8氮鸟嘌呤(8azaguanine,8 A G)从小鼠肺癌细胞 L A795 诱导抗8 A G 细胞株 A L A9702 ,并鉴定其生物学特性及成瘤性。结果 A L A9702 细胞在1.31×10- 4m ol/ L 8 A G 培养液中生长了1 年,在 H A T培养液中不能形成集落,并保持了来源细胞 L A795 的生长特性、染色体数目、成瘤性及肺转移性。 结论 A L A9702细胞保持了来源细胞的生物学性质及成瘤性质,是一株稳定的抗8 A G的小鼠肺癌细胞株,将作为适合细胞融合的抗原细胞,为研制肺癌疫苗,建立对肺癌有效的特异性免疫治疗奠定良好的基础。
Objective To develop a lung cancer vaccine by cell fusion, lung cancer antigen cells suitable for cell fusion need to be established. Methods A L A9702 cells were induced by 8azaguanine (8A G) from mouse lung cancer cell line L A795 and their biological characteristics and tumorigenicity were identified. Results A L A9702 cells grew for 1 year in 1.31×10-4 mol/L 8A G medium. Colonies could not be formed in H A T culture medium, and the growth characteristics of L A795 cells were preserved. Chromosomal number, tumorigenicity, and metastatic lung. Conclusion A L A9702 cell maintains the biological properties and tumorigenicity of the source cells. It is a stable anti-8-A G mouse lung cancer cell line, which will be used as an antigen-adapted cell fusion cell for the development of a lung cancer vaccine. It lays a good foundation for effective specific immunotherapy for lung cancer.
