目的:观察川红拔瘤膏对小鼠移植性肿瘤S180的抑制,探讨其诱导肿瘤细胞凋亡的作用。方法:建立小鼠荷瘤S180模型,随机分为5组,川红拔瘤膏按高、中、低3个剂量右腋下皮肤涂抹给药。模型组采用生理盐水灌胃给药,阳性组腹腔注射25mg/kg环磷酰胺。连续给药10d,观察瘤重、抑瘤率,DNA琼脂糖凝胶电泳和流式细胞仪器检测细胞凋亡与细胞周期变化。RT-PCR法检测凋亡基因Caspase-3的mRNA的表达。结果:与模型组比较,川红拔瘤膏高、中剂量能显著抑制小鼠S180的生长(P<0.05),可诱导肿瘤细胞凋亡,阻滞细胞周期在G0/G1期(P<0.05)。中、高剂量组均有Caspase-3表达的升高(P<0.05,P<0.01)。结论:川红拔瘤膏具有明显的抗肿瘤作用,作用机制可能是诱导凋亡,其诱导凋亡过程有Caspase-3的表达升高。
OBJECTIVE: To observe the inhibitory effect of Chuanhongbaopao cream on the transplanted tumor S180 in mice and to explore its role in inducing tumor cell apoptosis. Methods: The mouse model of S180 tumor-bearing mice was established and randomly divided into 5 groups. Chuanhongbaomai cream was applied to the right axillary skin at three doses of high, medium and low doses. The model group was given intragastric administration of saline, the positive group intraperitoneal injection of cyclophosphamide 25mg / kg. Continuous administration for 10 days, tumor weight, tumor inhibition rate, DNA agarose gel electrophoresis and flow cytometry were used to detect apoptosis and cell cycle changes. The mRNA expression of apoptosis gene Caspase-3 was detected by RT-PCR. Results: Compared with the model group, Chuanghongbaomai Oral Cream could significantly inhibit the growth of S180 mice (P <0.05), induce the apoptosis of tumor cells, and arrest the cell cycle in G0 / G1 phase (P <0.05) ). Caspase-3 expression increased in medium and high dose groups (P <0.05, P <0.01). CONCLUSION: Chuanhongbaomao cream has significant anti-tumor effect, and its mechanism may be apoptosis induction. Caspase-3 expression is increased in the process of apoptosis induction.
