目的:探讨细胞周期调控相关蛋白p53、p21、信号转导子和转录活化因子3(signal transducers and activators of transcription 3,STAT3)和细胞周期蛋白1(Cyclin D1)的表达在异常黑胆质型肝癌病证动物模型肝硬化期的发生发展中的作用.方法:根据维吾尔医学理论,在异常黑胆质载体大鼠模型的基础上,用二乙基亚硝胺(diethylnirtosamine)诱发建立维吾尔医异常黑胆质型肝癌病证模型肝脏标本用免疫组织化学法检测p53、p21、STAT3和Cyclin D1等基因的表达水平.结果:p53、p21、STAT3和Cyclin D1等基因的表达水平与对照组相比,对照肝癌组和异常黑胆质型肝癌病证组均上调,差异具有统计学意义(P<0.01);与对照肝癌组相比异常黑胆质型肝癌病证组上调,差异具有统计学意义(P<0.05).结论:异常黑胆质体液可能通过上调p53、p21、STAT3和Cyclin D1等基因的表达水平影响细胞周期调控,从而促进异常黑胆质型肝癌病证模型肝硬化的发生和发展.
Objective: To investigate the expression of p53, p21, signal transducers and activators of transcription 3 (STAT3) and cyclin D1 in abnormal savda hepatocellular carcinoma Methods: According to the Uighur medical theory, based on the abnormal savda carrier model in rats, diethylnitrosamine was used to induce the development of abnormal Uyghur black The expression of p53, p21, STAT3 and Cyclin D1 were detected by immunohistochemical staining in liver specimens of gallstone hepatocellular carcinoma.Results: Compared with the control group, the expression of p53, p21, STAT3 and Cyclin D1, Compared with control group, HCC group and abnormal savda hepatocarcinoma group were up-regulated, the difference was statistically significant (P <0.01); compared with the control group, the group of abnormal savda hepatocarcinoma increased, the difference was statistically significant ( P <0.05) .Conclusion: Aberrant sagittal body fluid may promote cell cycle dysregulation by up-regulating the expression of p53, p21, STAT3 and Cyclin D1 The occurrence and development model of cirrhosis.
