三苯氧胺(TAMOXIFEN,TXF)是临床乳腺癌内分泌治疗中重要的抗雌激素药物之一,长期用药可致耐药性的产生。体内体外实验模型的建立有助于阐明耐药性的机理。本文应用改良的Hug′S双层琼脂培养法,对SHZ-88细胞进行克隆培养,筛选出耐受TXF的大鼠乳腺癌细胞株。该细胞株对药物TXF的敏感性比原代细胞降低十倍,但对雌激素的反应性无改变。该细胞株的建立将有助于研究耐药乳腺肿瘤细胞的生物学行为,并可用于筛选抗耐药乳腺肿瘤药物。
Tamoxifen (TAMOXIFEN, TXF) is one of the important anti-estrogen drugs in clinical breast cancer endocrine therapy, long-term use of drugs can cause the emergence of drug resistance. The establishment of in vitro and in vivo experimental models helps elucidate the mechanism of drug resistance. In this paper, SHG-88 cells were cloned and cultured using a modified Hug’S double-layer agar culture method to screen TXF-resistant rat breast cancer cell lines. The sensitivity of this cell line to the drug TXF was ten times lower than that of the primary cells, but there was no change in estrogen reactivity. The establishment of this cell line will help to study the biological behavior of drug-resistant breast tumor cells, and can be used to screen anti-drug resistant breast tumor drugs.
