以硝普钠作为一氧化氮的供体。用流式细胞仪荧光强度测定、细胞ELISA、Northern杂交等方法分别从蛋白表达、基因表达水平研究一氧化氮对内皮细胞粘附分子VCAM-1表达的影响。结果表示:一氧化氮抑制IL-1α诱发的内皮细胞粘附分子VCAM-1的基因表达水平其作用里浓度及时间依赖性。一氧化氮抑制内皮细胞粘附分子VCAM-1的表达是其抑制单核一内皮细胞粘附及抗动脉粥样硬化的机制之一.
Sodium nitroprusside as a nitric oxide donor. The effects of nitric oxide on the expression of endothelial cell adhesion molecule VCAM-1 were studied by flow cytometry (FCM), cell ELISA and Northern blotting respectively from the protein expression and gene expression level. The results showed that nitric oxide inhibited IL-1α-induced endothelial cell adhesion molecule VCAM-1 gene expression in a concentration-dependent and time-dependent manner. Nitric oxide inhibits the expression of endothelial cell adhesion molecule VCAM-1 is one of the mechanisms of its inhibition of monocyte-endothelial cell adhesion and anti-atherosclerosis.
