目的探讨氯化钴预适应对大鼠局灶性脑缺血的保护作用及对nNOS基因表达的调节。方法建立线栓法大鼠局灶性脑缺血的动物模型,以30μg/kg氯化钴预处理大鼠,以TTC染色方法观察脑缺血范围,并以RT-PCR法检测nNOS基因的表达情况。结果氯化钴预适应能显著降低大鼠的脑梗塞面积,起到保护作用,并上调nNOS基因的表达,而nNOS阻滞剂7-NI可明显降低预适应的保护作用。结论氯化钴预适应可以对大鼠局灶性脑缺血起到保护作用,对nNOS基因的调节作用可能是其机制之一。
Objective To investigate the protective effect of cobalt chloride preconditioning on focal cerebral ischemia in rats and the regulation of nNOS gene expression. Methods The animal model of focal cerebral ischemia by thread occlusion was established. Rats were pretreated with 30μg / kg cobalt chloride. The extent of cerebral ischemia was observed by TTC staining. The expression of nNOS gene was detected by RT-PCR Happening. Results Cobalt chloride preconditioning could significantly reduce the area of cerebral infarction in rats and play a protective role and up-regulate the expression of nNOS gene, while nNOS blocker 7-NI can significantly reduce the protective effect of preconditioning. Conclusion Preconditioning of cobalt chloride can play a protective role in focal cerebral ischemia in rats. The regulation of nNOS gene may be one of its mechanisms.
