目的:通过利用HPLC监测羟乙桂胺乳膏离子导入与直接外敷在家兔的透皮吸收过程,为临床经皮给药途径的研究提供参考。方法:家兔背部脱毛后,一组离子导入负极给药;一组直接外敷给药,于给药前后各时间兔耳缘静脉采血1.5ml,预处理后HPLC检测其血药浓度。结果:药时曲线符合二室模型,离子导入组与直接外敷组cmax和tmax均有显著性差异(P<0.05),分别为2.578mg·ml-1,1.388mg·ml-1和1.146,1.608h。结论:离子导入有显著的促进透皮吸收效果
OBJECTIVE: To provide a reference for the clinical study of transdermal drug delivery in rabbits by iontophoresis and direct external application of HPLC. Methods: After the hair removal on the back of rabbits, a group of ions were introduced into the negative electrode. One group was administered directly by external application. 1.5 ml of blood was drawn from the rabbit’s ear vein before and after administration. The plasma concentration was detected by HPLC after pretreatment. Results: The curve of drugtime accorded with two-compartment model. There was significant difference between cmax and tmax in iontophoresis group and direct external application group (P <0.05), which were 2.578mg · ml-1 and 1.388mg · ml- 1 and 1.146,1.608h. Conclusion: Iontophoresis has a significant effect of promoting transdermal absorption
