目的:研究葫芦茶抗过敏性哮喘的活性组分及化学成分。方法:采用中、高压液相色谱法对葫芦茶叶提取物进行多组分富集,采用无标记整合药理学方法筛选富集得到的组分对GPR35受体的激动活性,应用HPLC-TOF-MS技术对活性组分的化学成分进行分析。结果:葫芦茶叶的95%乙醇提取物经正庚烷-70%甲醇萃取,70%甲醇萃取层经C18中压制备柱,分别以5%,70%和95%的甲醇洗脱,其中70%甲醇洗脱部分经XAmide高压制备色谱柱,以甲醇-水洗脱,按色谱峰收集流分,共得到10个组分。GPR35受体激动活性筛选结果显示组分Fr4,Fr5,Fr6,Fr7,Fr9,Fr10对GPR35受体有较强的激动活性,同时对GPR35受体激动剂敏喘宁显示了脱敏活性,由此推测上述组分为葫芦茶抗过敏性哮喘的活性组分。采用HPLC-TOF-MS技术对上述活性组分的化学成分进行了分析,从中鉴定出乌索酸,儿茶素,山柰酚-3-O-α-鼠李糖基(1→6)-β-葡萄糖苷,槲皮素-3-O-α-鼠李糖基(1→6)-β-葡萄糖苷/槲皮素-3-O-α-鼠李糖基(1→6)-β-半乳糖苷,间苯三酚-1-O-β-葡萄糖苷,槲皮素-3-O-β-葡萄糖苷,山柰酚-3-O-β-葡萄糖苷,山柰酚-3-O-β-半乳糖苷,triquetrumone E,triquetrumone F,山柰酚,胡萝卜苷,芹菜素,共12个化合物。结论:组分Fr4,Fr5,Fr6,Fr7,Fr9,Fr10为葫芦茶抗过敏性哮喘的有效组分,从活性组分中检测到的12个化合物可能为葫芦茶抗过敏性哮喘的有效成分。
Objective: To study the active components and chemical constituents of hyacinth tea against allergic asthma. Methods: The multi-component enrichment of gourd tea extract was carried out by medium and high pressure liquid chromatography. The excitatory activity of the enriched components on GPR35 receptor was screened by unlabeled integrated pharmacological method. The activity of GPR35 receptor was evaluated by HPLC-TOF-MS Technical Analysis of the chemical composition of the active ingredient. Results: 95% ethanol extract of gourd tea was extracted with n-heptane-70% methanol and 70% methanol was separated by 5%, 70% and 95% The methanol eluted part was subjected to high pressure XAmide preparative column to elute with methanol-water. The fractions were collected according to the chromatographic peaks, and a total of 10 fractions were obtained. GPR35 receptor agonist screening results showed that the components Fr4, Fr5, Fr6, Fr7, Fr9, Fr10 have strong agonistic activity on the GPR35 receptor, and desensitization activity to the GPR35 receptor agonist Minchuan Ning Speculated that the above components of the gourd tea anti-allergic asthma active component. The chemical constituents of the above mentioned active ingredients were analyzed by HPLC-TOF-MS. Ursolic acid, catechin, kaempferol-3-O-α-rhamnosyl β-glucoside, quercetin -3-O-α-rhamnosyl (1 → 6) -β-glucoside / quercetin -3-O- β -galactoside, phloroglucinol-1-O-β-glucoside, quercetin-3-O-β-glucoside, kaempferol-3-O-β-glucoside, 3-O-β-galactoside, triquetrumone E, triquetrumone F, kaempferol, daucosterol, apigenin, a total of 12 compounds. Conclusion: The components Fr4, Fr5, Fr6, Fr7, Fr9 and Fr10 are the effective components of gourd tea against allergic asthma. The 12 compounds detected from the active components may be the effective components of gourd tea anti-allergic asthma.
