目的分析丙型肝炎病毒(HCV)RNA分型试剂盒检测深圳市抗-HCV阳性献血者结果。方法收集2014-2015年158份深圳市抗-HCV阳性献血者血液样本,应用聚合酶链反应(PCR)-荧光探针法进行HCV RNA定量检测,病毒载量>1.0×103 IU/mL的样本经HCV RNA分型试剂盒检测HCV基因型,分析不同基因型所占比例,病毒基因型与载量之间的相关性。结果 158份抗-HCV阳性献血者PCR-荧光探针法检出HCV RNA阳性样本54例,病毒载量>1.0×103 IU/mL的45例,全部得到分型结果,HCV 1b型、2型、3型、6型分别占57.78%(26/45)、6.67%(3/45)、8.89%(4/45)、26.67%(12/45)。单因素方差分析结果显示,1b型与2型病毒载量差异有统计学意义(F=2.861,P<0.05);不同性别间HCV RNA定量检测结果及抗-HCV S/CO值结果差异有统计学意义(P<0.05);不同年龄段各基因型分布比例经Fisher精确检验,差异有统计学意义(P<0.05)。结论 HCV 1b型、6型仍为深圳市无偿献血人群感染HCV的主要基因型,而HCV 2型和3型比例有所减少。
Objective To analyze the result of detecting anti-HCV positive blood donors in Shenzhen with hepatitis C virus RNA typing kit. Methods A total of 158 anti-HCV positive blood samples from Shenzhen were collected from 2014 to 2015. HCV RNA was detected by polymerase chain reaction (PCR) -Fluorescent probe method. The samples with viral load> 1.0 × 103 IU / mL The HCV genotypes were detected by HCV RNA typing kit, and the correlation between different genotypes and viral genotypes was analyzed. RESULTS: Totally 54 positive samples of HCV RNA were detected by PCR-fluorescence probe method in 158 anti-HCV positive donors, and 45 cases with viral load> 1.0 × 103 IU / mL were screened. All the HCV genotypes 1b and 2 , Type 3 and type 6 accounted for 57.78% (26/45), 6.67% (3/45), 8.89% (4/45) and 26.67% (12/45), respectively. The results of one-way ANOVA showed that there was a significant difference in the viral load between type 1b and type 2 (F = 2.861, P <0.05). There was statistical difference between the quantitative results of HCV RNA and the value of anti-HCV S / CO (P <0.05). The distribution of genotypes in different age groups was statistically significant (P <0.05) by Fisher’s exact test. Conclusion The HCV genotypes 1b and 6 are still the major HCV genotypes in Shenzhen donation blood donors, while the proportion of HCV genotypes 2 and 3 is reduced.
