目的观察益气化瘀解毒法对慢性萎缩性胃炎伴异型增生模型大鼠P21WAF1和Cylin D1基因的影响。方法将60只Wistar大鼠随机选取50只造模,其余10只为空白组。造模成功后将大鼠随机分为模型组、维酶素组、消痞颗粒组,每组各11只。模型组予生理盐水,维酶素组予维酶素悬浊液,消痞颗粒组予消痞颗粒中药制备药液,1次/d。连续治疗12周后用实时荧光定量PCR法(RT-PCR)及Western blot法对各组大鼠P21WAF1和细胞周期素D1(Cyclin D1)基因进行检测。结果P21WAF1mRNA及蛋白表达量消痞颗粒组较模型组及维酶素组显著增高(P<0.05)。Cyclin D1mRNA及蛋白表达量消痞颗粒组较模型组、维酶素组显著降低(P<0.05)。结论中医益气化瘀解毒法可以逆转胃黏膜异型增生,这种作用可能是通过促进P21WAF1的表达、抑制Cyclin D1的表达,从而恢复P21-CDK-Cyclin D1之间的平衡,进而使细胞周期调控恢复正常而实现的。
Objective To observe the effects of Yiqi Huayu detoxification method on P21WAF1 and Cylin D1 gene in chronic atrophic gastritis with dysplasia rats. Methods 60 Wistar rats were randomly selected 50 models, the remaining 10 were blank. After successful modeling, the rats were randomly divided into model group, Vitexin group and Xiaopi granule group, with 11 rats in each group. The rats in the model group were treated with normal saline, and the vitamin D group was treated with Vitexin suspension, Xiaopi granule group was given Xiaopi granule Chinese medicine to prepare the liquid, once / d. After continuous treatment for 12 weeks, the gene P21WAF1 and Cyclin D1 in each group were detected by real-time fluorescence quantitative PCR (RT-PCR) and Western blot. Results The expression of P21WAF1 mRNA and protein in Xiaopi granule group was significantly higher than that in model group and Vitexin group (P <0.05). The expression of Cyclin D1 mRNA and protein in Xiaopi granule group was significantly lower than that in model group and verapamil group (P <0.05). Conclusion The method of clearing away qi and removing blood stasis with detoxification can reverse the dysplasia of gastric mucosa. This effect may restrain the expression of P21WAF1 and the expression of Cyclin D1, thus restoring the balance between P21-CDK-Cyclin D1 and further regulating cell cycle Return to normal and achieve.
