采用典型的籼粳交(ZYQ8/JX17)的花药培养DH群体为材料,在15℃的条件下对水稻发芽力进行研究.发现该 DH群体的籼、粳双亲在低温条件下的发芽力存在明显差异,6~11d,ZYQ8的发芽率比JX17高;12~16 d,JX17的发芽率比ZYQ8高.用该群体构建的分子连锁图谱进行了发芽力逐日数量性状座位(QTL)分析,共检测到2个QTL,其中位于第9染色体C397B和RZ617B之间的qLTG-9在8~11d检测到其加性效应均为正值,即来自JX17的等位基因有利于提高发芽力;位于第4染色体RG908和CT563之间的qLTG-4在12~16d检测到其加性效应均为负值,即ZYQ8的等位基因提高发芽力.2个QTL表现时间不同,显示了低温条件下发芽力遗传机制的复杂性.
The DH population of typical populations of indica and japonica hybrids (ZYQ8 / JX17) was used as material to study the germination ability of rice at 15 ℃. The results showed that there was a significant difference in germination ability between indica and japonica parents at low temperature in this DH population. The germination rate of ZYQ8 was higher than that of JX17 from 6 to 11 days. The germination rate of JX17 was higher than that of ZYQ8 from 12 to 16 days. Two QTLs were detected using QTL analysis of the molecular linkage map constructed by this population, of which qLTG-9 located on the 9th chromosome C397B and RZ617B detected their QTLs at 8 ~ 11d The genotypes of JX17 were positive for improving the germinability. The additive effect of qLTG-4 located between chromosomes RG908 and CT563 on chromosome 4 was negative at 12-16 days, which was negative for ZYQ8 The allele increases germination. The two QTLs showed different timing, showing the complexity of the genetic mechanism of germination in low temperature.
