目的观察中药淫羊藿、女贞子对地塞米松干预大鼠哮喘模型全身及局部不同水平糖皮质激素受体(GR)的影响。方法建立大鼠哮喘模型,腹腔注射地塞米松2周治疗,同时配合灌胃淫羊藿女贞子煎液,流式细胞术检测支气管肺泡灌洗液(BALF)与血淋巴细胞中GR蛋白及GR结合力的表达。结果哮喘模型组血淋巴细胞及BALF中GR结合力均显著降低(P<0.05或P<0.01);与哮喘模型组比较,激素干预组BALF中GR蛋白表达量明显降低(P<0.01),血淋巴细胞及BALF中GR结合力均明显降低(P<0.05或P<0.01);应用激素的同时合用淫羊藿女贞子,能显著上调BALF中GR蛋白量(P<0.05)及血淋巴细胞及BALF中GR结合力(P<0.05或P<0.01)。结论在激素应用的同时合用中药淫羊藿、女贞子在整体及局部水平均能提高因糖皮质激素(GC)干预下降的GC结合力,改善GR对GC的耐受,提高支气管肺组织及机体对激素的敏感性。
Objective To observe the effects of Herba Epimedii and Fructus Ligustri Lucidi on dexamethasone at different levels of systemic and local glucocorticoid receptor (GR) in asthmatic rats. Methods The rat model of asthma was established by intraperitoneal injection of dexamethasone for 2 weeks. At the same time, the effects of Ligustrum lucidum and Fructus Ligustri Lucidi decoction were observed. The expressions of GR protein in bronchoalveolar lavage fluid (BALF) and blood lymphocytes were detected by flow cytometry and Expression of GR binding. Results Compared with the asthma model group, the expression of GR protein in the BALF of the asthma model group was significantly decreased (P <0.01), while that of the BALF was significantly lower in the asthma model group (P <0.05 or P <0.01) (P <0.05 or P <0.01). The combined use of hormones with Epimedium acinar Ligustrum lucidum could significantly upregulate the GR protein level in BALF (P <0.05) and the increase in blood lymphocytes And GR binding in BALF (P <0.05 or P <0.01). Conclusion The combination of hormones with Chinese Herba Epimedii and Fructus Ligustri Lucidi can improve the GC binding ability of GC due to decreased glucocorticoid (GC) intervention, improve the tolerance of GC to GC, and improve the bronchial lung tissue and The body’s sensitivity to hormones.
