目的:研究巴戟天正丁醇提物对乳鼠心肌细胞缺氧/复氧损伤(H/R)的防护作用。方法:采用培养的乳鼠心肌细胞建立缺氧/复氧损伤模型,实验分正常细胞培养组、缺氧/复氧损伤模型组、巴戟天醇提物小、中、大剂量组。观察心肌细胞形态并测定活性氧(ROS)、内皮素(ET-1)的含量变化。结果:巴戟天醇提物可明显改善缺氧/复氧损伤的心肌细胞形态及搏动程度;降低ROS、ET-1的含量。结论:巴戟天正丁醇提物对缺氧/复氧损伤的心肌细胞有防护作用;其防护作用可能与降低细胞液中ROS、ET-1的含量有关。
Objective: To study the protective effect of n-butanol extracts of Morinda officinalis against hypoxia / reoxygenation injury (H / R) in neonatal rat cardiomyocytes. Methods: Hypoxia / reoxygenation injury model was established by cultured neonatal rat cardiomyocytes. The experiment was divided into normal cell culture group, hypoxia / reoxygenation injury model group, small, medium and large dose of Morinda officinalis alcohol extract. The morphology of cardiomyocytes was observed and the contents of reactive oxygen species (ROS) and endothelin (ET-1) were measured. Results: Ethanol extract of Morinda officinalis could significantly improve the cardiomyocyte morphology and pulsatility in hypoxia / reoxygenation injury and decrease the content of ROS and ET-1. CONCLUSION: The ethanol extract of Morinda officinalis has a protective effect on cardiomyocytes injured by hypoxia / reoxygenation. Its protective effect may be related to the decrease of ROS and ET-1 in the cytosol.
