目的:研究复方藤梨根制剂逆转人红白血病耐药细胞株K562/ADM裸鼠移植瘤的多药耐药效应及机制。方法:建立K562/ADM裸鼠移植瘤多药耐药模型;以流式细胞仪(FCM)检测各组肿瘤细胞膜上P-gp的表达和肿瘤细胞内ADM蓄积浓度。结果:(1)ADM组P-gp的表达与NS组相比明显升高(P(0.05);不同剂量的FFTLG制剂与ADM合用时,P-gp的表达与ADM组相比显著下降(P(0.01)。单独应用FFTLG制剂,当浓度为0.8mg/mL时P-gp的表达与ADM组相比下降(P(0.05)。(2)与ADM组相比,ADM和不同剂量的FFTLG合用组肿瘤细胞内的ADM浓度均有上升(P(0.05)。结论:FFTLG制剂可以部分逆转荷瘤裸鼠的多药耐药性,其逆转效果似呈剂量依赖性。FFTLG制剂逆转荷瘤裸鼠多药耐药性的机理之一是通过下调肿瘤细胞膜上P-gp的表达或抑制P-gp的活性,增加肿瘤细胞内ADM的蓄积浓度而实现的。
Objective: To study the multidrug resistance effect and mechanism of compound ramie root preparations in reversing the transplanted tumor of human erythroleukemia cell line K562 / ADM. Methods: The multidrug resistance model of K562 / ADM xenografts in nude mice was established. The expression of P-gp and the accumulation of ADM in tumor cells were detected by flow cytometry (FCM). Results: (1) The expression of P-gp in ADM group was significantly higher than that in NS group (P <0.05). The P-gp expression was significantly decreased in different dose of FFTLG preparation compared with ADM group (P (P <0.05). (2) Compared with the ADM group, ADM and different doses of FFTLG combined with the application of the FFTLG preparation, the P-gp expression decreased (P (0.05)) .Conclusion: FFTLG preparation can partially reverse multidrug resistance of tumor-bearing nude mice, and the reversal effect seems to be dose-dependent.FTFG preparation can reverse the tumor-bearing nude mice One of the mechanisms of multidrug resistance is through down-regulating the expression of P-gp in tumor cell membrane or inhibiting the activity of P-gp and increasing the accumulation concentration of ADM in tumor cells.
