去七肽(B24-30)胰岛素(DHPI)是胰岛素B链C端经限制性酶解法切去相应肽段后没有活力但能结晶的胰岛素类似物。DHPI能结晶,因此用X-光衍射法研究DHPI的立体结构,对于了解胰岛素的结构和功能的关系有重要作用。前此报道DHPI的晶体是薄片状的,因此进一步寻找了其它的结晶。结果发现,去七肽胰岛素可以在含某些有机溶剂的0.05M柠檬酸溶液中结晶。结晶方法基本上用过饱和静止法。先配DHPI的贮存液,使含有0.05
De-Heptapeptide (B24-30) Insulin (DHPI) is a non-viable but crystallizable insulin analog that is cleaved off the C-terminus of the insulin B chain by restriction enzyme digestion. DHPI can crystallize, so the study of the three-dimensional structure of DHPI by X-ray diffractometry plays an important role in understanding the relationship between structure and function of insulin. The previously reported crystals of DHPI are flaky, so further crystallizations are sought. As a result, it has been found that the decapheotide insulin can be crystallized in a 0.05 M citric acid solution containing some organic solvent.
