目的探讨麻黄素对小鼠肺组织的影响。方法 60只小鼠随机分为麻黄素组和对照组,采用递增剂量腹腔连续注射麻黄素溶液(0.0315 g/kg、0.0477 g/kg、0.0564 g/kg)和等量生理盐水15d(每天两次,每次0.2ml),用比色法检测各组小鼠肺组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量及血浆髓过氧化物酶(MPO)活性的变化,用苏木素-伊红染色法观察肺组织结构的变化,用免疫组织化学法检测肺组织c-Fos蛋白和Caspase-3蛋白表达的变化。结果麻黄素组小鼠肺组织SOD、CAT活性5d时暂时升高,10d、15d时显著降低,MDA含量5d时暂时降低,10d、15d时显著升高;血浆MPO活性显著升高,肺组织病理损伤明显,肺泡直径和肺泡隔厚度显著增大,放射状肺泡数(RAC)明显减小;肺组织c-Fos、Caspase-3蛋白阳性表达的平均吸光度值明显增强。结论麻黄素可加重机体炎症反应,影响小鼠肺组织抗氧化酶活性,诱导肺组织细胞凋亡,导致小鼠肺的组织结构和功能受损。
Objective To investigate the effect of ephedrine on the lung tissue of mice. Methods Sixty mice were randomly divided into ephedrine group and control group. The mice were injected intraperitoneally with escalating doses of ephedrine solution (0.0315 g / kg, 0.0477 g / kg, 0.0564 g / kg) and normal saline for 15 days , 0.2ml each time). The contents of superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) in lung tissue and the levels of plasma myeloperoxidase (MPO). The changes of lung tissue structure were observed by hematoxylin-eosin staining. The expression of c-Fos protein and Caspase-3 protein in lung tissue were detected by immunohistochemical method. Results The activity of SOD and CAT in lungs of ephedrine group increased temporarily on the 5th day, decreased significantly on the 10th and 15th day, while decreased on the 5th day and increased on the 10th and 15th day. The activity of MPO in plasma increased significantly, The diameter of alveoli and the thickness of alveolar septum significantly increased, while the radial alveolar number (RAC) decreased obviously. The average absorbance of c-Fos and Caspase-3 protein in lung tissue was significantly increased. Conclusion Ephedrine can aggravate the inflammatory reaction in the body, affect the activity of antioxidant enzymes in the lung tissue of mice and induce the apoptosis of lung cells, leading to the impaired tissue and function of lung in mice.
