目的探讨HBV相关性和非HBV相关性肝细胞癌(HCC)组织的长链非编码RNA(lncRNA)的表达情况。方法选择行手术治疗的HBV相关HCC患者42例、非HBV相关HCC患者45例,采用原位杂交技术检测两组患者肿瘤维修组和癌旁组织中肌动蛋白丝相关蛋白1-反义RNA1(AFAP1-AS1)、肝癌微血管浸润相关lncRNA(MVIH)和肺腺癌转移相关转录本1(MALAT-1)的表达水平,并比较不同临床特征HCC患者肿瘤组织AFAP1-AS1、MVIH和MALAT-1的表达情况。结果在两组患者中,肿瘤组织AFAP-AS1的染色分数低于癌旁组织(P<0.05),而肿瘤组织MVIH和MALAT-1的染色分数高于癌旁组织(P均<0.05)。HBV相关性HCC组癌旁组织中的AFAP-AS1染色分数高于非HBV相关HCC组癌旁组织(P<0.05),而两组肿瘤组织AFAP-AS1染色分数、两组肿瘤组织或癌旁组织中MVIH和MALAT-1的染色分数比较,差异均无统计学意义(P均>0.05)。在HBV相关性HCC、非HBV相关性HCC患者中,AFAP1-AS1、MVIH和MALAT-1在不同的分期和分型患者的肿瘤组织中的表达水平比较,差异均无统计学意义(P均>0.05)。结论在HCC组织中AFAP1-AS1表达下调而MVIH和MALAT-1表达上调,三者均参与了HCC的发生发展,而AFAP1-AS1的表达可能还受到HBV相关生物学过程的调节。
Objective To investigate the expression of long chain non-coding RNA (lncRNA) in HBV-related and non-HBV-associated hepatocellular carcinoma (HCC) tissues. Methods Forty-two patients with HBV-related HCC and 45 patients without HBV-related HCC undergoing surgical treatment were selected. In situ hybridization was used to detect the expression of actin-related protein 1-antisense RNA1 AFAP1-AS1, MVIH and MALAT-1 in HCC, and compared the expression of AFAP1-AS1, MVIH and MALAT-1 in HCC patients with different clinical features Express the situation. Results In both groups, the staining score of AFAP-AS1 in tumor tissue was lower than that in paracancer tissue (P <0.05), while the staining scores of MVIH and MALAT-1 in tumor tissue were higher than those in paracancerous tissue (all P <0.05). The staining score of AFAP-AS1 in the paratumor tissue of HBV-related HCC group was higher than that in non-HBV-related HCC group (P <0.05), while AFAP-AS1 staining scores of the two groups of tumor tissues or paracancerous tissues There was no significant difference in the staining scores between MVIH and MALAT-1 (all P> 0.05). There were no significant differences in the expression levels of AFAP1-AS1, MVIH and MALAT-1 between the HBV-related HCC and non-HBV-related HCC patients in different stages and subtypes of patients (P> 0.05). Conclusions The expression of AFAP1-AS1 is down-regulated and MVIH and MALAT-1 are upregulated in HCC tissues, all of which are involved in the development of HCC. The expression of AFAP1-AS1 may be regulated by HBV-related biological processes.
