目的了解外周血淋巴细胞染色体标本制片的影响因素,寻找出最佳的室内质量控制制作条件。方法比较制片过程中不同的接种血量、培养温度、低渗时间以及离心速度对外周血中染色体标本制作的影响情况。结果外周血染色体制片过程中最佳的室内质量控制条件是接种血量0.7×32规格注射器针头20~25滴;培养温度应该严格控制在37℃±0.5℃;最佳低渗时间是15~20 min;最佳离心速度1 500 r/min~2 000 r/min;固定液应在使用前临时配置,放置1 h应废弃;玻片冷藏处理的效果比热水处理后的好。结论外周血淋巴细胞染色体标本制片的室内质量控制条件有接种血量、培养温度和低渗时间等,相关实验室应予关注,对辐射防护作出正确的评价。
Objective To understand the influencing factors of peripheral blood lymphocyte chromosome specimen preparation and to find out the best conditions for the production of indoor quality control. Methods The effects of different inoculated blood volume, culture temperature, hypotonic time and centrifuge speed on the production of chromosome samples in peripheral blood during the process of preparation were compared. Results The best quality control conditions in peripheral blood chromosomal preparation were 20-25 drops of inoculum of 0.7 × 32 gauge syringe; the culture temperature should be strictly controlled at 37 ℃ ± 0.5 ℃; the optimal time of hypotonicity was 15 ~ 20 min; the optimal centrifugal speed is 1 500 r / min ~ 2000 r / min; the fixative solution should be temporarily configured before use and should be disposed for 1 h; the effect of slide refrigeration treatment is better than that after hot water treatment. Conclusion Laboratory quality control of peripheral blood lymphocyte chromosome preparation should be concerned with the blood volume, culture temperature and hypotonic time, and the radiation protection should be evaluated correctly.
