目的 :评价拓扑替康对人新鲜乳腺癌细胞的抗癌效果 ,并将其与 5种乳腺癌常用化疗药物对人新鲜乳腺癌细胞的抑制作用进行对比。方法 :以 2 0例已确诊的新鲜乳腺癌癌块为实验对象进行人新鲜乳腺癌细胞培养药物敏感性试验。结果 :(1)人乳腺癌细胞原代培养结果 :2 0例乳腺癌标本中 ,阳性率 85 % (17/ 2 0 ) ,获得 1× 10 5/ m L的乳腺癌单细胞悬液 10 m L以上。 (2 )原代培养人乳腺癌细胞对 6种抗癌药物敏感性试验结果 :常规浓度组敏感率依次为 :PTX>ADM>TPT>MTX>5 - Fu>DDP。 (3)统计学分析χ2 检验 ,TPT高、中浓度组之间差异无显著性 (P>0 .0 5 ) ,两者与低浓度组之间差异有显著性 (P<0 .0 5 )。结论 :(1)拓扑替康对乳腺癌有效 ,相当于氨甲蝶呤和 5 -氟尿嘧啶的作用。 (2 )乳腺癌细胞培养药物敏感性试验作为临床筛选新的抗乳腺癌药物的方法之一 ,敏感性较高 ,有进一步推广应用的价值
OBJECTIVE: To evaluate the anti-cancer effect of topotecan on human breast cancer cells and to compare the inhibitory effects of topotecan on human breast cancer cells and the five commonly used chemotherapy drugs for breast cancer. Methods: Twenty cases of newly diagnosed breast cancer were used as experimental subjects for drug sensitivity test of human breast cancer cell culture. Results: (1) The results of primary culture of human breast cancer cells: The positive rate was 85% (17/20) in 20 breast cancer samples and the single cell suspension of 1 × 10 5 / m L was obtained 10 m L or more. (2) The results of sensitivity test of primary cultured human breast cancer cells against six kinds of anticancer drugs: The sensitivity of routine concentration group was: PTX> ADM> TPT> MTX> 5 - Fu> DDP. (3) Statistical analysis There was no significant difference between the high and medium TPT groups (χ2 test) (P> 0.05), the difference between the two groups was significant (P <0.05) . Conclusion: (1) Topotecan is effective in breast cancer, which is equivalent to the effect of methotrexate and 5 - fluorouracil. (2) Breast cancer cell culture drug sensitivity test As one of the clinical screening methods for new anti-breast cancer drugs, the sensitivity is high, so it has the value of further popularization and application
