自杀基因前药系统是目前肿瘤基因治疗中的研究热点。其中CD/5-FC自杀基因前药系统是目前研究最多、进展最快的自杀基因系统之一。它对肿瘤细胞的杀死机制除了通过直接毒性作用和旁观者效应以外,还包括自杀基因联合使用、免疫细胞和免疫调节因子的参与、自杀基因与放射治疗联合作用等。本文就CD/5-F自杀基因前药系统的分子生物学基础、抗癌机制以及其在临床实验中面临的问题等研究进展进行综述。
Suicide gene prodrug system is currently a hot research topic in gene therapy of cancer. One CD / 5-FC suicide gene prodrug system is currently the most studied and the fastest growing suicide gene system. Its mechanism of killing tumor cells in addition to through the direct toxicity and bystander effect, including the joint use of suicide genes, immune cells and immune regulatory factors involved in suicide gene and radiotherapy combined effect. This review summarizes the molecular biology of the CD / 5-F suicide gene prodrug system, its anticancer mechanism and the problems it faces in clinical trials.
