阿糖腺甙(adenine arabinoside,ARA-A)是1960年由Lee等首次用人工合成的核甙,在体外、内均证明对DNA病毒有较广泛、显著的抗病毒作用。早期临床上主要用于局部治疗单纯疱疹病毒和带状疱疹病毒感染,70年代初发现静脉滴注ARA-A可明显降低单纯疱疹病毒或巨细胞病毒所致脑炎的病死率。1978年Pollard等试用于治疗慢性乙型肝炎,证明此药对乙型肝炎病毒(HBV)有明显抑制作用。1984年我国首次合成ARA-A获得成功,现正进行临床验证之中。
Adenosine arabinoside (ARA-A) was the first artificial nucleoside synthesized by Lee et al in 1960, and has been proved to have a broad and significant antiviral effect against DNA viruses both in vitro and in vivo. Early clinical mainly for the local treatment of herpes simplex virus and herpes zoster virus infection, early 70s found that intravenous infusion of ARA-A can significantly reduce herpes simplex virus or cytomegalovirus-induced encephalitis mortality. In 1978, Pollard and other trials for the treatment of chronic hepatitis B, the drug proved to have significant inhibitory effect on hepatitis B virus (HBV). In 1984, China’s first synthetic ARA-A was successful, and clinical validation is underway.
