目的:探讨ETS1/2对非洲爪蛙胚胎胰腺发育的影响。方法:设计并合成特异性抑制ETS1/2表达的反义吗啉代寡核苷酸(morpholino oligonueleitide,MO),通过显微注射的方法将MO转入爪蛙胚胎,利用整胚原位杂交和定量RT-PCR检测标志性基因的表达变化。结果:单独敲降ETS1或ETS2对爪蛙胚胎发育无明显影响,共同敲降ETS1/2后会使爪蛙胚胎发育异常,胰腺标志基因表达上升,肠道标志基因表达下降,肝脏和胃标志基因无明显变化;胰腺标志基因ngn3、igf1、foxa1的启动子区存在ETS1/2的结合位点。结论:单独敲降ETS1或ETS2对爪蛙胚胎发育无明显影响,敲降ETS1/2促进爪蛙胚胎胰腺发育,抑制肠道发育,对肝脏和胃的发育无明显影响。
Objective: To investigate the effect of ETS1 / 2 on pancreas development in Xenopus laevis. Methods: Antisense morpholino oligonucleotides (MOs) were designed and synthesized to specifically inhibit the expression of ETS1 / 2. The MOs were transformed into clawed frog embryos by microinjection. Quantitative RT-PCR detection of gene expression changes. RESULTS: ETS1 or ETS2 alone had no significant effect on the development of clawed frog embryo. Combined knockdown of ETS1 / 2 resulted in the abnormal development of clawed frog embryo, increased expression of pancreatic marker gene, decreased expression of intestinal marker gene, decreased expression of liver and gastric marker genes No obvious changes were observed. The pancreatic marker genes ngn3, igf1 and foxa1 had ETS1 / 2 binding sites. CONCLUSION: ETS1 or ETS2 knocked down alone had no significant effect on the development of clawed frog embryos. Knockdown of ETS1 / 2 promoted the development of pancreatic clawed frog embryonic pancreas, inhibited the development of the intestinal tract and had no significant effect on the development of the liver and stomach.
