目的 Dishevelled(Dvl)亚家族成员在非小细胞肺癌中均存在过表达,但他们与β-catenin和p120ctn及患者预后的关系尚不清楚。方法采用S-P免疫组化和RT-PCR法检测80例非小细胞肺癌(NSCLC)组织中Dvl-1、Dvl-3与β-catenin、p120ctn表达的关系及对预后的影响。结果正常支气管黏膜上皮Dvl-1、3均呈阴性表达,而在肺癌组织中表达上调,其异常表达率分别为52.5%(42/80)和51.3%(41/80),肺腺癌的阳性率均高于鳞癌,并与NSCLC的低分化高分期和淋巴结转移有关(<0.05)。Dvl-1高表达与β-catenin异常表达相关(<0.01),而Dvl-3高表达与p120ctn异常表达相关(<0.01)。Dvl-1、3的异常表达与非小细胞肺癌患者不良预后呈正相关(<0.05)。结论 NSCLC组织中存在着Dvl-1和Dvl-3的过表达,并与组织学分型、分化、临床分期和淋巴结转移相关。Dvl-1、3过表达分别与β-catenin、p120ctn异常表达相关。Dvl-1、3在非小细胞肺癌组织的异常表达可作为评估预后的辅助指标。
The purpose of the Dishevelled (Dvl) subfamily members are over-expressed in non-small cell lung cancer, but their relationship with the prognosis of β-catenin and p120ctn is not yet clear. Methods The relationship between the expression of Dvl-1, Dvl-3, β-catenin and p120ctn in 80 cases of non-small cell lung cancer (NSCLC) and their prognosis was detected by S-P immunohistochemistry and RT- Results The normal expression of Dvl-1,3 in bronchial mucosa epithelium was negative, while it was up-regulated in lung cancer tissues. The abnormal expression rates were 52.5% (42/80) and 51.3% (41/80) respectively. The positive rates of lung adenocarcinoma The rates were higher than that of squamous cell carcinoma, and correlated with poorly differentiated staging and lymph node metastasis of NSCLC (<0.05). The high expression of Dvl-1 was correlated with the abnormal expression of β-catenin (<0.01), while the high expression of Dvl-3 was correlated with the abnormal expression of p120ctn (<0.01). The abnormal expression of Dvl-1,3 was positively correlated with the poor prognosis of non-small cell lung cancer (P <0.05). Conclusion Overexpression of Dvl-1 and Dvl-3 exists in NSCLC tissues, which is associated with histological classification, differentiation, clinical stage and lymph node metastasis. Dvl-1, 3 overexpression and β-catenin, p120ctn abnormal expression. Abnormal expression of Dvl-1,3 in non-small cell lung cancer can be used as an adjunct to assess prognosis.
