目的探讨高浓度葡萄糖对大鼠视网膜Müller细胞超微结构和低氧诱导因子-1α(HIF-1α)表达的影响。方法体外传代培养大鼠视网膜Müller细胞,分正常对照组、葡萄糖25 mmol/L组和50 mmol/L组,电镜观察不同组Müller细胞超微结构的改变,W estern印迹检测不同组Müller细胞中HIF-1α的表达。结果葡萄糖25 mmol/L组Müller细胞超微结构发生了明显的改变:细胞核缩小、不规则,核內染色质呈凝集边聚;胞质浓染,线粒体肿胀,可见空泡结构。葡萄糖50 mmol/L组,胞质内可见残余体,细胞表面绒毛减少变钝。W estern印迹结果示正常对照组无HIF-1α蛋白的表达,葡萄糖25 mmol/L组和50 mmol/L组均有低氧诱导因子-1α蛋白的表达,50 mmol/L组蛋白表达量低于25 mmol/L组。结论高浓度葡萄糖可以引起Müller细胞超微结构的改变,并引起HIF-1α的表达。
Objective To investigate the effects of high glucose on the ultrastructure of retinal Müller cells and the expression of hypoxia inducible factor-1α (HIF-1α) in rats. Methods The rat retinal Müller cells were subcultured and cultured in vitro. The changes of ultrastructure of Müller cells in different groups were observed by electron microscopy in normal control group, 25 mmol / L glucose group and 50 mmol / L glucose group. Western blotting was used to detect HIF -1α expression. Results The ultrastructure of Müller cells in 25 mmol / L glucose group changed obviously: the nucleus was reduced and irregular, the chromatin in nucleus was aggregated and aggregated; the cytoplasm was dense and the mitochondria were swollen and the vacuole structure was observed. Glucose 50 mmol / L group, visible in the cytoplasm remnants of the cell surface villi reduce dull. Western blotting results showed that there was no expression of HIF-1α protein in normal control group. The expression of hypoxia inducible factor-1α protein in 25 mmol / L glucose group and 50 mmol / L glucose group was lower than that in 50 mmol / L group 25 mmol / L group. Conclusion High concentrations of glucose can induce ultrastructural changes in Müller cells and induce the expression of HIF-1α.
