目的研究人工麝香对金黄色葡萄球菌的生长代谢的影响,人工麝香质量生物活性评价新方法。方法采用微量热法,测定37℃时金黄色葡萄球菌在不同浓度的人工麝香作用下的热谱曲线,并获得其相应的生长速率常数(k)、最大发热功率(Pm)及其出现时间(tm)、总发热量(Qt)等热动力学参数,经主成分分析和回归分析等化学计量学方法综合评价人工麝香对金黄色葡萄球菌生长代谢的影响。结果人工麝香对金黄色葡萄球菌生长代谢具有明显的抑制作用,这种抑制作用主要受第一、二指数生长期最大发热功率出现时间(tm1、tm2)的影响。结论通过分析这两个主要参数的数值变化,能更方便、快捷和准确地评价人工麝香抑制金黄色葡萄球菌活性,为进一步评价人工麝香及其他药物的抗菌生物活性提供了有用的方法和基础。
Objective To study the effect of artificial musk on the growth and metabolism of Staphylococcus aureus and a new method for evaluating the biological activity of artificial musk quality. Methods The thermogram of Staphylococcus aureus under different concentrations of artificial musk was determined by microcalorimetry at 37 ℃. The corresponding growth rate constants (k), maximum heating power (Pm) and their appearance time ( tm), total heat (Qt) and other thermodynamic parameters. The effects of artificial musk on the growth and metabolism of Staphylococcus aureus were comprehensively evaluated by stoichiometric methods such as principal component analysis and regression analysis. Results Artificial musk had a significant inhibitory effect on the growth and metabolism of Staphylococcus aureus. The inhibitory effect was mainly influenced by the tm1 and tm2 of the maximum heating power in the first and second exponential growth phase. Conclusion By analyzing the numerical changes of these two main parameters, it is more convenient, quick and accurate to evaluate the activity of artificial musk against Staphylococcus aureus and provide a useful method and basis for further evaluating the antibacterial biological activity of artificial musk and other drugs.
