目的研究(E)-苯乙基-3-(3,5-二羟基-4-异丙基苯基)丙烯酸酯(THCA354)化合物的抗炎作用及抗炎初步机制。方法通过二甲苯和佛波酯诱导的小鼠耳肿胀模型来评价THCA354的抗炎作用;进一步通过佛波酯诱导的小鼠耳肿胀模型来评价该化合物对小鼠耳组织中髓过氧化酶(MPO)的活性和IL-1β、IL-6、TNF-α炎症细胞因子的抑制作用。结果与模型组相比,局部涂抹THCA354化合物的小鼠耳朵厚度,淋巴细胞的浸润,耳组织中髓过氧化物酶活性和IL-1β、IL-6、TNF-α炎症细胞因子的表达均显著降低。结论 THCA354化合物可能通过抑制中性粒细胞的活化和IL-1β、IL-6、TNF-α炎症细胞因子的释放从而有效地缓解小鼠耳组织炎症水肿和淋巴细胞的浸润,发挥抗炎作用。
Objective To investigate the anti-inflammatory and anti-inflammatory mechanism of (E) -phenylethyl-3- (3,5-dihydroxy-4-isopropylphenyl) acrylate (THCA354) Methods The anti-inflammatory effect of THCA354 was evaluated by mouse ear edema model induced by xylene and phorbol ester. Furthermore, the effect of the compound on myeloperoxidase MPO) activity and IL-1β, IL-6, TNF-α inhibitory effect of inflammatory cytokines. Results Compared with the model group, the thickness of the ear, the infiltration of lymphocytes, the myeloperoxidase activity and the expression of IL-1β, IL-6 and TNF-α inflammatory cytokines were significantly increased in the THCA354-treated mice reduce. Conclusions THCA354 compounds may exert anti-inflammatory effects by inhibiting the activation of neutrophils and the release of inflammatory cytokines such as IL-1β, IL-6 and TNF-α, thereby effectively relieving inflammation and edema in the ear tissues of mice and infiltration of lymphocytes.
