目的 探讨原癌基因蛋白质 C- erb B- 2及增殖细胞核抗原 (PCNA )在结肠癌中含量及与结肠癌生物学行为的关系。 方法 利用免疫组织化学及自动化图像分析技术定量检测 C- erb B- 2及 PCNA在 10例正常肠粘膜 ,30例结肠腺瘤 ,5 3例结肠癌中的表达。 结果 阳性总面积、积分光密度、平均光密度、阳性率 1 、平均光密度 1及半定量测定的阳性率等 6个参数值均按正常肠粘膜→ 级不典型增生腺瘤→ 级不典型增生腺瘤→ 级不典型增生腺瘤→腺癌的顺序呈递变趋势 ,伴有淋巴结转移的腺癌组中 ,上述各参数值明显增高。 结论 C- erb B- 2及 PCNA一起可作为判断肿瘤的恶性程度及评价预后的指标 ,应用图像分析技术对结肠癌及癌前病变进行多指标多参数的计量分析 ,具有鉴别诊断的重要意义 ,为结肠癌的早期诊断提供客观依据。
Objective To investigate the relationship between the expression of proto-oncogene C-erb B-2 and proliferating cell nuclear antigen (PCNA) in colon cancer and the biological behavior of colon cancer. Methods Immunohistochemistry and automated image analysis were used to quantitatively detect the expression of C-erb B-2 and PCNA in 10 cases of normal intestinal mucosa, 30 cases of colon adenoma and 53 cases of colon cancer. Results Six parameters of positive total area, integral optical density, average optical density, positive rate 1, average optical density 1 and semi-quantitative determination of positive rate were all in the order of normal intestinal mucosa → atypical hyperplastic adenoma → atypical hyperplasia Adenoma → grade atypical hyperplastic adenoma → adenocarcinoma showed a trend of change in order, with lymph node metastasis of adenocarcinoma group, the above parameters were significantly higher. Conclusions C erb B-2 and PCNA together can be used as indicators to judge the malignant degree of tumor and evaluate the prognosis. Using image analysis technique to measure multiple parameters and multiple parameters of colon cancer and precancerous lesions has the important significance of differential diagnosis. To provide an objective basis for the early diagnosis of colon cancer.
