目的:观察水通道蛋白1(AQP1)在重症急性胰腺炎(SAP)大鼠肺组织中的表达,从新的角度探讨肺损伤(ALI)发生的机制。并观察清胰汤对SAP大鼠肺脏AQP1表达的影响。方法:Wistar大鼠制备SAP模型,随机分为4组:假手术对照组(SHAM组)、SAP模型组(SAP组)、SAP+清胰汤治疗组(QYD组)、SAP+地塞米松治疗组(DEX组)。各组又分为6、12、24 h组,分别于造模后6、12、24 h处死,取肺组织及动静脉血。检测血清淀粉酶水平(AMY),测定动脉PaO_2、Pa CO2。取肺脏计算肺湿干重比值(W/D)、大体及镜下观察肺脏病理学改变,并用免疫组化和RT-PCR方法测定肺脏水通道蛋白1蛋白及m RNA的表达情况。结果:与SHAM组相比,SAP组AMY、Pa CO2、W/D值明显升高(P<0.01),PaO_2值明显下降,AQP1 m RNA和AQP 1蛋白表达明显下降(P<0.01)。与SAP组相比,QYD治疗组AMY、Pa CO2、W/D值明显下降(P<0.01),PaO_2值明显升高(P<0.01),AQP1 m RNA和AQP 1蛋白表达明显升高(P<0.01)。与SAP组相比,DEX治疗组AQP1 m RNA和AQP 1蛋白表达明显升高(P<0.01)。DEX治疗组和QYD治疗组之间AQP1 m RNA和AQP 1蛋白表达无显著差异(P>0.05)。结论:水通道蛋白1表达与急性胰腺炎损伤关系密切,清胰汤可上调其表达而减轻肺水肿。
Objective: To observe the expression of aquaporin 1 (AQP1) in the lungs of rats with severe acute pancreatitis (SAP) and explore the mechanism of lung injury (ALI) from a new perspective. The effect of Qingyi Decoction on AQP1 expression in the lungs of SAP rats was observed. Methods: SAP model was prepared in Wistar rats and divided into 4 groups at random: sham operation control group (SHAM group), SAP model group (SAP group), SAP + Qingyiyoup treatment group (QYD group), SAP + dexamethasone treatment group DEX group). Each group was divided into 6,12,24 h group, were killed 6,12,24 h after modeling, lung tissue and arteriovenous blood. Serum amylase level (AMY) was measured, arterial PaO_2 and PaCO_2 were measured. Lung wet / dry weight ratio (W / D) was taken from the lungs. Lung pathological changes were observed under the microscope and under microscope. The expression of aquaporin 1 and m RNA in the lungs were detected by immunohistochemistry and RT-PCR. Results: Compared with SHAM group, the AMY, PaCO 2 and W / D values in SAP group were significantly increased (P <0.01), PaO_2 values were significantly decreased, and the expression of AQP1 m RNA and AQP 1 protein were significantly decreased in SAP group (P <0.01). Compared with SAP group, the levels of AMY, PaCO 2 and W / D in QYD treatment group decreased significantly (P <0.01), PaO_2 level increased significantly (P <0.01), AQP1 m RNA and AQP 1 protein expression increased <0.01). Compared with SAP group, the expression of AQP1 m RNA and AQP1 protein in DEX treatment group was significantly increased (P <0.01). There was no significant difference in the expression of AQP1m RNA and AQP1 between DEX treatment group and QYD treatment group (P> 0.05). Conclusion: The expression of aquaporin-1 is closely related to the injury of acute pancreatitis. Qingyitang can up-regulate its expression and reduce pulmonary edema.
