目的建立一种简单有效的人晶状体上皮细胞体外培养的方法。方法用组织块培养法,对不同年龄组(儿童组16例,中青年组18例,老年组14例)的人晶状体前囊膜进行培养。对培养的细胞进行形态学和增殖活动观察。结果组织块培养法较易在体外培养人类晶状体上皮细胞并传代,其增殖能力与供体年龄有关,但是老年人晶状体上皮细胞无法传代。结论成功地建立起人晶状体上皮细胞体外培养模型,可用于白内障及后发性白内障时后囊膜混浊发病机制和药物试验研究。
Objective To establish a simple and effective method of in vitro culture of human lens epithelial cells. Methods Tissue explant culture was used to culture the anterior lens capsule of different age groups (16 in children group, 18 in middle-aged group, and 14 in aged group). The cultured cells were observed for morphological and proliferative activity. Results Tissue mass culture was easier to culture human lens epithelial cells in vitro and passaged. Its proliferative ability was related to donor age, but the lens epithelial cells in the elderly could not be passaged. Conclusion The successful establishment of human lens epithelial cells culture model in vitro can be used for cataract and posterior capsular opacification pathogenesis and drug experimental study.
