目的 测定肝脏移植患者CsA血药浓度。方法 :采用一步萃取法提取 ,高效液相色谱法测定。色谱柱 :phenomenexC1 8(2 5 0mm× 4.6mm ,5um) ;流动相 :CH3OH :H2 O =85 :15 ;柱温 :73℃ ;流速 :1.0ml min ;检测波长 :2 14nm。结果 CsA的保留时间为 9.0min ;定量线性范围为 2 0 -60 0ng(S N≥ 3 ) ,方法回收率 (10 0 .0 8± 1.3 8) % (n =5 ) ,日内、日间RSD <10 % (n =5 )。结论 :本方法快速 ,定量准确 ,可用于血药浓度常规监测及人体药代动力学研究。
Objective To determine the CsA concentration in liver transplantation patients. Methods: The extraction was performed by one-step extraction and determined by high performance liquid chromatography. Column: phenomenexC1 8 (250 mm × 4.6 mm, 5 μm); mobile phase: CH 3 OH: H 2 O = 85:15; column temperature: 73 ° C .; flow rate: 1.0 ml min .; detection wavelength: 214 nm. Results The retention time of CsA was 9.0 min. The quantitative linear range was 20-6060ng (SN≥3). The recovery rate of the method was (10.08 ± 1.3 8)% (n = 5) 10% (n = 5). Conclusion: The method is rapid, accurate and accurate and can be used for routine monitoring of blood concentration and pharmacokinetics of human body.
