目的:研究天麻素对Ox-LDL诱导牛主动脉内皮细胞(BAECs)黏附HL-60细胞的抑制作用及其机制。方法:通过细胞黏附实验观察天麻素对BAECs与HL-60细胞的黏附作用;流式细胞术检测ICAM-1和VCAM-1的表达;细胞色素C还原法测定细胞含量;报告基因法检测核转录因子κB(NF-κB)转录活性的变化。结果:天麻素可使Ox-LDL诱导的BAECs黏附HL-60细胞显著降低,BAECs细胞表面ICAM-1和VCAM-1表达下降,细胞含量以及NF-κB转录活性也显著下降。结论:天麻素能抑制Ox-LDL诱导的BAECs黏附HL-60细胞。
Objective: To investigate the inhibitory effect of gastrodin on the adhesion of HL-60 cells induced by Ox-LDL in bovine aortic endothelial cells (BAECs) and its mechanism. Methods: The adhesion of gastrodin to BAECs and HL-60 cells was observed by cell adhesion assay. The expression of ICAM-1 and VCAM-1 was detected by flow cytometry. The content of cells was measured by cytochrome C reduction assay. Change of NF-κB transcriptional activity. Results: Gastrodin could significantly reduce the adhesion of HL-60 cells induced by Ox-LDL to BAECs. The expression of ICAM-1 and VCAM-1 on the surface of BAECs decreased, and the content of cells and the transcriptional activity of NF-κB decreased significantly. Conclusion Gastrodin can inhibit the adhesion of HL-60 cells induced by Ox-LDL to BAECs.
