目的:探讨中药复方更年汤对肾阳虚型围绝经期模型大鼠肾上腺皮质中Bax、Bcl-2表达的影响。方法:建立自然衰老的雌性肾阳虚型围绝经期大鼠模型,11~12月龄60只,随机分为更年汤组、利维爱组、生理盐水组,每组20只;4月龄20只作为青年组;18月龄20只作为老年组。用免疫组化法检测肾上腺皮质Bax、Bcl-2的表达。结果:与生理盐数组比较,利维爱组肾上腺皮质Bax表达降低、Bcl-2的表达升高,差异均有统计学意义(P<0.05)。更年汤组肾上腺皮质Bax阳性总积分表达低于利维爱组、生理盐水组,Bcl-2阳性总积分表达高于利维爱组、生理盐水组,差异均有统计学意义(P<0.05),更年汤组稍优于利维爱组。结论:自拟更年汤能降低肾阳虚型围绝经期模型大鼠肾上腺皮质Bax的表达,提高肾上腺皮质Bcl-2表达水平,平衡围绝经期内分泌失调。
Objective: To investigate the effect of Gengnian decoction on the expression of Bax and Bcl-2 in the adrenal cortex of rats with perimenopausal model of Shen Yang Deficiency. Methods: To establish a model of spontaneous aging female rats with kidney-yang-deficiency syndrome. Sixty children, 11-12 months old, were randomly divided into three groups, namely, the Gengnian decoction group, the levofloxacin group and the saline group. 20 as young group and 20 as old group. Immunohistochemistry was used to detect the expression of Bax and Bcl-2 in adrenal cortex. Results: Compared with the physiological saline group, the expression of Bax and the expression of Bcl-2 in adrenal cortex decreased in Levi’s group, with statistical significance (P <0.05). The positive expression of Bax in adrenal cortex of Gengnian Decoction group was lower than that of Leviosin group (P <0.05), and the positive expression of Bcl-2 in total ginsenoside group was significantly higher than that of Levi group and saline group ), Gengnian decoction group was slightly better than Levi group. Conclusion: Gengnian decoction can reduce the expression of Bax in the adrenal cortex of rats with perimenopausal model of deficiency of the yin deficiency, increase the expression of Bcl-2 in the adrenal cortex and balance the imbalance of peri-menopause.
