目的研究肿瘤坏死因子(TNFα)对溶血卵磷脂(LPC)引起脑微血管平滑肌细胞(SMC)增殖的影响及维生素E(VitE),普罗布考(PRB)的保护作用。方法用结晶紫染色测定密度(OD)值的方法观察TNFα对LPC引起牛脑微血管平滑肌细胞(BCMSMC)增殖作用的影响。结果TNFα在25×104~1×105U·L-1范围内能协同LPC引起BCMSMC增殖,8h时增殖率就明显增加,16h增殖率达峰值(672%)。VitE20,50,100μmol·L-1,PRB10,20,50μmol·L-1呈剂量依赖性地显著降低其增殖率,其最大抑制率分别为742%和497%。结论TNFα对LPC引起的BCMSMC增殖有协同作用,药物VitE,PRB可拮抗其增殖作用。
Objective To study the effect of tumor necrosis factor α (TNFα) on the proliferation of cerebral microvascular smooth muscle cells (SMC) induced by lysophosphatidylcholine (LPC) and the protective effect of vitamin E and probucol (PRB). Methods The method of crystal violet staining determination of density (OD) was used to observe the effects of TNFα on the proliferation of bovine cerebral microvascular smooth muscle cells (BCMSMC) induced by LPC. Results TNF α could promote the proliferation of BCMSMC with LPC in the range of 2.5 × 104-1 × 105U · L-1. The proliferation rate increased significantly at 8h and reached the peak at 16h (672%). VitE20,50,100μmol·L-1, PRB10,20,50μmol·L-1 significantly reduced the proliferation rate in a dose-dependent manner, with the maximum inhibition rates of 742% and 497%, respectively. Conclusion TNF @ LPC-induced proliferation of BCMSMC have a synergistic effect, the drug VitE, PRB can antagonize its proliferation.
